Rafael Nadal's bid for a record 21st Grand Slam title came crashing to a halt at the Australian Open on Wednesday as Stefanos Tsitsipas rose up to topple the Spaniard 3-6, 2-6, 7-6(4), 6-4, 7-5 and reach the semi-finals.

World number two Nadal had not lost a set coming into the evening clash at Rod Laver Arena and was locked on target for the last four at Melbourne Park after roaring to a two-set lead over the listless Greek.

The match turned on its head when Nadal wavered in a messy third set tiebreak, and a rejuvenated Tsitsipas rallied brilliantly to take the match into a decider.

Capturing Nadal's serve at 5-5 in the fifth, Tsitsipas saw two match points slip through his fingers as he served for the match but fired a backhand winner down the line on the third to claim one of his finest career victories.

"Well done for him, he played better than me probably in the important moments," Nadal told reporters.

"I tried my best in every single moment ... I think I stayed positive all the time during the match, fighting, and it was not enough. Sometimes it's enough, today was not enough."

Fifth seed Tsitsipas will meet Russian Daniil Medvedev for a place in the final.

"I fly like a little bird and everything was working for me," Tsitsipas said on court.

"The emotions in the end are indescribable."

Russian Daniil Medvedev secured a spot in the Australian Open semi-finals with a 7-5, 6-3, 6-2 victory over friend and compatriot Andrey Rublev on Wednesday, extending his winning streak to 19 matches.

It was Rublev's fourth straight defeat to Medvedev, who has not lost a match since October.

Fourth seed Medvedev had won all his previous matches against Rublev in straight sets and it was no different at a scorching Rod Laver Arena on Wednesday.

ATP Finals winner Medvedev raised his game during the important points and sealed the contest with a forehand winner to set up a last-four meeting against either second-seeded Spaniard Rafa Nadal or fifth seed Stefanos Tsitsipas of Greece.

Russian qualifier Aslan Karatsev will meet world number one Novak Djokovic in the other semi-final on Thursday, with the prospect of an all-Russian men's final looming on Sunday.

World No 1 Ashleigh Barty bowed out of the Australian Open after losing to Czech Karolina Muchova on Wednesday.

Muchova defeated the local hero 6-1, 3-6, 2-6 in a match that lasted for 117 minutes on Rod Laver Arena. The win sends the 25th seed, Muchova into her first Grand Slam semi-final.

Muchova didn't have a good start as she lost the first set and took a medical timeout in the second set having her temperature checked. But she made a remarkable comeback winning the last two sets 6-3, 6-2.

Jennifer Brady battled through her frustrations to overhaul friend and United States team mate Jessica Pegula 4-6, 6-2, 6-1 and reach her maiden Australian Open semi-final on Wednesday.

Brady, the 22nd seed, was a picture of anguish early in the clash on a stifling day at Rod Laver Arena but gradually dialled in her power game before crushing Pegula in the final set.

Brady reached her second Grand Slam semi-final after making the last four at last year's US Open.

With world number one Ash Barty shocked by Karolina Muchova in the early quarter-final on Wednesday, Brady has a golden chance to reach her maiden Slam final.
